基本転写因子の欠損による遺伝病の病態と機能の関連性に対する生化学及び遺伝学的解析

研究概要	1.線虫TFIIEを用いたPolIIの転写開始から伸長への移行段階の解析我々は、TFIIEの機能を解析する目的で線虫TFIIEを単離しヒトTFIIEと比較検討した。ヒトと線虫TFIIEのサブユニットを組み合わせたキメラTFIIEを発現・精製し、転写再構成系にてヒトTFIIEと機能的互換性を検討したところ、線虫βはヒトβと部分的に置き換ったが、線虫αは置き換らなかった。原因を検討すると、線虫αはヒトβとの結合活性が10%以下となりヒトTFIIEの様な転写活性を示さないことが分った。一方、線虫βはヒト転写系では伸長への移行活性が低かった。TFIIEは、PolII最大サブユニットのCTD配列の2番目と5番目セリンのTFIIHによるリン酸化を促進する。ヒトαと線虫βのキメラでCTDリン酸化を調べると、5番目セリンリン酸化活性が著しく低下していた。このことから今回、伸長への移行はPolIICTDの5番目セリンリン酸化と密接な関連性があることが示唆された。 2.TFIIHを欠損する遺伝病患者細胞の欠損TFIIHのDNA修復反応における他因子との協調的制御機能の解析 TFIIHはPolIIの転写に必要であるが、一方、紫外線等によるDNA傷害修復にも直接関与し、9サブユニットの2個XPBとXPDのどちらか一方に変異が入ることで発がんを伴う色素性乾皮症(XP)が発症する。我々は、これまでにTFIIHが遺伝子DNA上の紫外線、薬剤等による傷害部位にDNA修復因子が集まり修復する際に、形成される複合体内で損傷を認識することを明らかにしたXPCと損傷DNA部位の3'側を切断するXPGの2者に結合し機能することを見出している。今回、これら因子が実際にヒト細胞内でTFIIHと相互作用し、その際紫外線、薬剤により損傷を生じたDNAとの相互作用の効率は、損傷の無いものより高いことを明らかにした。
