研究概要

アラキドン酸カスケードにより生合成されるプロスタグランジン(PG)類は生体の恒常性の維持に重要な役割を果たすとともに、その過剰産生は炎症、アレルギー等の病変の一因となることが知られている。本代謝系に関与する酵素としてsPLA2-IIA及びcPLA2、COX-1及びCOX-2が知られており、これまでの申請者の研究により、様々な細胞と刺激の組み合わせによりPG合成に利用されるPLA2とCOXのカップリングが異なることがわかってきた。また、試験管内ではアラキドン酸選択性を示さないsPLA2-IIAが細胞内ではアラキドン酸を選択的に遊離することも観察している。これらの現象を考察するとPLA2とCOXの機能を制御する細胞因子が存在することが想定された。細胞内でcPLA2と結合するタンパク質をfar-Western法で検索した結果、細胞骨格系の中間径フィラメント構成タンパク質であるビメンチンが同定された。本結合はカルシウム依存的であり、ビメンチンが核辺縁部に豊富に存在することから、各種細胞で観察されるカルシウム濃度上昇に伴うcPLA2の細胞質から核辺縁部への細胞内トランスロケーションに本結合が関与することが予想された。MAPキナーゼによるリン酸化でcPLA2が活性化されることが知られている。本研究ではカルシウムにより制御を受けるCaMKIIに注目し、その特異的阻害剤であるKN-93をラット線維芽細胞に前処理した結果、カルシウムイオノフォア刺激に伴うPGE2産生が顕著に抑制された。このときErk1およびp38 MAPキナーゼの活性化にKN-93は影響しなかったことから、MAPキナーゼを介さない経路でcPLA2が活性化される可能性が示唆された。
また、sPLA2-IIAはヘパラン硫酸を結合したGPIアンカータンパク質であるグリピカンと結合し、細胞表面のカベオラに集積することを見いだした。さらに、サイトカイン刺激細細胞で核周辺部にsPLA2-IIAが検出されることから、グリピカンはsPLA2-IIAの細胞内移行に関与することが予想された。
