新規プロテインキナーゼSIKのcAMP依存性核外移行の分子機構

研究概要	申請者が最近クローニングした塩誘導性キナーゼ(SIK)はN末端側にキナーゼドメインを有する新規のセリン/トレオニン・プロテインキナーゼである。これまでの実験の結果、非刺激時の副腎皮質細胞ではSIKは核内に存在するが、ACTHで刺激した細胞ではSIKが細胞質に存在すること、そしてSIKの細胞内移動がステロイドジェネシスの開始に重要であることが分かった。GFPを融合したSIKを作成してSIKの細胞内局在を詳細に検索した。その結果、静止状態のY1細胞ではSIKは核内に存在するが、Y1をACTHで刺激するとSIKは核から細胞質に出る。この移動は5分以内に完了する。核外への移動はレプトマイシンで阻害される。SIKの核外移行にはPKAが関与する。PKA活性を欠いたY1変異株(Kin-7)では観察されないが、PKA発現ベクターを共導入すると核外移行が起こる。PKAによってSIKのSer577がリン酸化されることがSIKの核外移行に重要である。そしてACTH刺激後のSIKの細胞内移動の時間経過は、SIKによるステロイド産生酵素遺伝子の発現抑制作用の時間経過を良く反映する。細胞質で生合成されたSIKタンパク質が細胞質から核に移行するのにはC末端側に位置する塩基性アミノ酸を比較的多く含む50アミノ酸残基が必要である。これらの結果は、ステロイドジェネシスのシグナル伝達機構の調節にSIKが細胞内移動を介してエッセンシャルな役割を演ずることを示唆している。
