癌抑制遺伝子p53の新規類似遺伝子の検索およびクローニング

研究概要	最近、私が確立した縮重プライマーを用いたPCR(degenerate primer PCR(DPP)法)は、類似遺伝子が存在すれば必ずクローニングが可能といっても過言ではないほど強力である。p51、p73の単離に引き続き、同定した新p53類似遺伝子p53Rの解析を行うことで本研究費を申請したが、この遺伝子は偽遺伝子であることが判明した。したがって、p53ファミリー遺伝子の解析全般を行うことに方針を変更し、以下の点を明らかにした。 p51とp73はp53に構造上、機能上の類似性を有する。これらの遺伝子について以下の解析結果を得た。(1)キメラ遺伝子を用いた解析-キメラ遺伝子の機能の多様性を利用すべくアデノウイルス組み換え体を作製し、遺伝子治療、p53ファミリー遺伝子の機能解析における有効性を示した。(2)p51、p73のゲノム解析-両ゲノム配列のほぼ全長を決定し、両者はゲノム上も類似性が高いことを明らかにし、プロモーター領域の解析を行った。(3)新規アイソフォームの解析-新規アイソフォームをΔNp73は、TAp73に対する結合阻害、p53に対する競合阻害活性を有すること見いだした。(4)p51、p73蛋白質の制御機構解析-TAp51はp53の主たる制御蛋白質MDM2には制御されないが、顕著なユビキチン化による制御を受けていることを見いだした。(5)DNp51の機能-DNp51BはIKKαによりリン酸化、分解を受け角化細胞の分化を制御していることを見いだした。
