環境汚染による疾病感受性の個人差を薬理遺伝学的に立証するための分析疾学的検討

研究概要

目的:遺伝薬理学と疫学の専門家による国際共同研究が、環境化学物質に起因する疾患の発生機序と遺伝因子の関係を解析するために有用であり、日本のmolecular epidemiologyの発展にも役立つと考えて本企画調査を行った。
実績:4回のカナダ訪問と、各施設での資料収集、意見交換によって、1)サスカチュアン州を中心とした特定地域における、住民の健康、農業活動、環境汚染(特に農薬)に関する医学・化学・農学・地質水質学者による共同研究(Prairie Ecosystem Study)のデーター集積状況を確認した。2)バングラデシュにおける砒素汚染については、現在住民の戸籍作成の段階であり、本研究実施には時期尚早であると判断した。3)近年のgene-environment interactionに関するmolecular epidemiologyの論文を、遺伝薬理学と疫学の立場から収集・評価して、今後の研究計画を立案した。
今後の展開:サスカチュアン州において本研究を実施するために、1)研究分担者の中川がサスカチュアン大学医学部のAdjective Professorに申請し、検体収集と解析を実行する留学生派遣を準備している。2)本企画を具体化して2001年のカナダ研究助成金に申請する予定である。3)その研究内容は、これまでサスカチュアン州で継続されてきた農薬曝露による気管支喘息・前立腺癌発症に関するコホート研究に、疾患(農薬毒)感受性遺伝因子としての薬物代謝酵素(Cytochrome P450(CYP)1A2,CYP3A4,CYP17,CYP19,N-acetyltransferase 2,Glutathione-S-transferase P1,Paraoxonase)の遺伝的多型調査を組み込んで今後2年間で多変量解析を行うものである。その後には、さらに焦点を絞った前向き調査も計画している。
