低分子量G蛋白質Rhoの情報伝達と生理的意義の研究

研究概要	Rhoとそのエフェクター(ROCK, mDiaなど)について以下の実験を行った。 (1)mDiaによるアクチン束の配列機構を解析し、これがアクチンのみならず、そのFH2領域を介して微小管をも配向させること、これにより細胞接着斑を形成しストレスファイバーの配列を決定していること、を見出した。また、この機構が外部張力による細胞接着斑形成のメカノセンサーとして働いていることを示した。(2)Y-27632とC3酵素の作用を比較することで、mDiaの情報伝達経路を解析し、これがCas-Crk-DOCKを経てRacの活性化に繋がること、ROCK経路はこの経路を抑制することを示し、RhoからRacに至る経路の存在とRho下流の二つの情報伝達経路の相互作用を明らかにした。(3)ROCK-IIの遺伝子欠損マウスを作製し、ホモ欠損マウスの殆どが胎生期に死亡すること、これが胎盤機能不全によることを明らかにした。(4)ROCKがcaspaseによって切断されて活性化されること、これがアポトーシスのさいのBleb形成に働いていることを示した。(5)Rho活性化因子ECT-2の結合蛋白質としてCIN-85を見出し、これらが分裂時に複合体を形成し分裂溝に局在することを示した。(6)細胞質分裂時のRhoエフェクターの局在を検討し、mDiaがFH3領域のC端を介して紡錘体微小管に局在することを見出した。また、Citron-kinaseの分裂に伴う動員機構を解析し、これがRho依存性に細胞質から分裂溝に移行すること、Rhoを不活化するとこの移行が中心紡錘体で停止することを見出した。(7)Pull-down法を用いてRho関連蛋白質の一つCdc42の分裂時の活性化を解析し、これがmetaphaseで一過性に上昇することを見出し、変異体の発現実験でこの上昇もその後の下降のどちらも正常分裂のため必要であることを見出した。
