SH3ドメインをもつアダプター蛋白質の活性化と高次構造変化のメカニズム

研究概要	食細胞NADPHオキシダーゼの酵素本体は、細胞膜のシトクロムb_<558>である。本酵素の活性化には、特異的アダプター蛋白質(p47^<phox>,p67^<phox>とp40^<phox>:各々SH3ドメインをもつ)が刺激依存性に細胞質から細胞膜に移行してシトクロムb_<558>と相互作用する必要がある。私共はオキシダーゼの活性化機構を研究し、平成13年度は以下のような成果を得た。 (1)p40^<phox>は休止時細胞でp67^<phox>と会合している蛋白質であるが、この両者の結合は、新規なドメイン間(p40^<phox>のPCモチーフとp67^<phox>のPB1ドメイン)の全く新しいタイプの蛋白質間相互作用によるものであることを示した。更に、これら新規なドメインが酵母のシグナル伝達蛋白質(Cdc24pとBem1p)にも存在し、酵母のシグナル伝達においても重要な役割を担うことも明らかにするとともに、Bem1pのPB1ドメインの3次構造決定に成功した。p40^<phox>の役割は今まで不明だったが、PB1-PC相互作用によるp67^<phox>との結合を介してp67^<phox>とp47^<phox>の膜移行を促進させ、オキシダーゼ活性化を正に制御していることを明らかにした。 (2)p47^<phox>とp40^<phox>(オキシダーゼの調節因子)のN末領域に新規ドメインを見い出していたが(PX/PB2ドメイン)、更に、P47^<phox>PXドメインの3次構造を決定するとともに、PXドメインがphosphoinositides結合能をもつこと、P47^<phox>のPXドメインはp47^<phox>の膜移行およびオキシダーゼ活性化に必須であることを示した。またp47^<phox>のPXドメインがSH3ドメインとの分子内結合により負に制御されていることを明らかにした。
