HIVアクセサリー蛋白質Vpx/Vprの機能解析

研究概要	HIV-2に特異的に存在するアクセサリー蛋白質であるVpxおよびVpxに構造的に類似するVprのT細胞中での機能につきプロウイルスクローンの欠損体や各種マーカークローンを用いて系統的に解析した。VpxがマクロファージでのウイルスDNAの核移行に必須であることは知られていたが、T細胞での機能は不明であり、また、Vprのウイルス複製に関する役割はほとんど報告がなかった。 Vpx欠損体はサルのT細胞であるHSC-Fで野性株よりはるかに増殖が悪く、VpxがT細胞でのウイルス複製に重要であることが明らかになった。Vpr欠損体には複製欠損が認められなかったが、Vpx/Vpr2重欠損体はVpx欠損体よりさらに増殖が遅れた。Vpx/Vpr欠損体の複製欠損性は他のヒト株化細胞では見られなかった。各種クローンを用いた定量的解析の結果、Vpx欠損体の複製欠損部位はウイルスDNAの核移行段階にあることが示された。Vpx/Vpr2重変異体はこれに加えてウイルス粒子の集合/放出段階にも欠損があった。また、Vpx/Vpr欠損体はヒト末梢リンパ球でHSC-F細胞中と同様の複製欠損性を示した。ヒトマクロファージでのウイルス複製にはVpxが必須でありVprは不必要であることも確認された。これらの成績は、(1)HIV-2DNAの核移行過程にはT細胞でもマクロファージでもVpxが関与しVprは関わっていないこと、(2)HIV-2Vprのウイルス粒子の集合/放出過程における機能はVpxが存在しない時にのみ明らかになること、さらに、(3)T細胞やマクロファージのような自然宿主細胞でのHIV-2複製にはVpxとVprが極めて重要な役割を果たすこと、を示している。
