HIV-1の吸着と侵入機構の解析

研究概要	通常HIV-1は細胞表面のCD4とケモカインレセプターであるCXCR4あるいはCCR5をレセプターペアとして、その感染が成立する。この感染は、さらに、糖脂質などの物質で細胞を前処理すると増強され、これらの物質が補助的に作用していることが報告されている。しかし、その詳しい機序はわかっていない。植物由来の糖脂質であるカプシアノサイドGは、細胞への前処理で、HIV-1のX4株のみの感染を選択的に増強した。また、X4R5株のウイルスではCXCR4とCD4を発現した細胞をカプシアノサイドGで処理することによって、その感染が増幅された。共焦点顕微鏡を用いた観察で、カプシアノサイドG処理した細胞ではCD4とCXCR4とのcappingと共集合が見られ、CD4とCCR5との間ではそのような現象は認められなかった。FITCでラベルしたHIV-1(X4株)では、ウイルス吸着後にFITCのcappingが観察されたが、その細胞の頻度は低かった。カプシアノサイドによるX4 HIV-1の感染増強は、この糖脂質によってCD4とCXCR4とが細胞の一部分に集められ、ウイルスの吸着と侵入が促進されたためと思われた。この現象とHIV-1レセプターを多く発現している細胞がHIV-1感染に感受性が高いという事実を考え合わせると、HIV-1は一個のウイルスが多くのレセプターとくっ付くことにより細胞内への侵入すると推測された(multiple site binding)。このmultiple site bindingは細胞膜の流動性が高くなるとその多結合の機会が高くなると仮定されている。この仮説を証明するため、吸着時の温度を40℃に設定した。この高温処理によりHIV-1の感染は増強された。今後、multiple site bindingを阻害する因子(抗体の附着など)を追究し、ウイルスの吸着と侵入の機構を明らかにしたい。
