植物水チャネルとCa^<2+>輸送システムの細胞・器官特異性と多細胞系構築

研究概要

(1)液胞膜Ca^<2+>/H^+対向輸送体と液胞膜Ca^<2+>結合タンパク質の器官特異性液胞へのCa^<2+>集積は植物細胞普遍的な現象と考えられるが、この機能を支える分子のいくつかは明瞭な器官特異性を示すことが明らかになった。今年度、液胞膜Ca^<2+>/H^+対向輸送体を対象に、イネから3種のcDNA(OsCAX1、OsCAX2、OsCX3)を単離し、Ca^<2+>輸送活性を酵母発現系で確認した。このうちCa^<2+>選択性の高いOsCAX1についてイネの根と葉でタンパク質量を比べると、葉で顕著に多く、しかも光照射で酵素量が減少した。液胞へのCa^<2+>集積システムが器官により異なるものと推定している。
一方、ダイコンでは、液胞のCa^<2+>結合タンパク質(RVCaB45)の解析を進めた。このタンパク質は1分子あたり19個のCa^<2+>を結合する。ダイコンをCa^<2+>十分な条件で生育するとRVCaB45発現量が抑制され、Ca^<2+>を欠く条件では発現が著しく誘導されることを見出した。RVCaB45は根、塊根での発現量が多く、葉ではほとんど検出されない。このことも液胞へのCa^<2+>集積機構が器官により異なることを示している。
(2)液胞膜、細胞膜水チャネルの植物ホルモン応答性 水チャネルの役割は、組織生長時にみられる細胞内、液胞内への水の取込み、浸透圧調節に関わる自由な水の輸送、細胞を横断する組織全体での水の流れの確保である。対象としているダイコンには細胞膜水チャネルとして2グループ(PAQ1型、PAQ2型)があり、水チャネル蓄積の細胞特異性(細胞膜水チャネルでは、実生根の中心柱と内皮、生長塊根では篩部で高発現)と水ストレスおよび植物ホルモンへの応答性(PAQ2型分子種のみが変動)を明らかにした。液胞膜型はとちらかといえば、恒常的にどの組織でも一定レベル以上発現した。以上のように水チャネルが、構成的発現グループと、環境応答型グループに2分されることを見出した。
