高等植物・茎の光屈性現象を制御する鍵化学物質

研究概要	光屈性は植物が具備する環境応答反応の代表的な生物機能である。従来、光屈性は光側と影側組織におけるオーキシンの偏差分布によって引き起こされるというCholodny-Went説によって説明されてきた。しかし、機器分析によるオーキシン量の測定から、オーキシンの偏差分布は全く起こらないことが明らかになり、更に光側組織で成長抑制物質が生成し、光側の成長が抑制されることによって屈曲することが明らかにされている(Bruinsma-Hasegawa説)。本研究は光屈性を制御する成長抑制物質(鍵化学物質)を探索し、更にその生成機構と作用機構を明らかにすること等を主な目的として研究を行った。様々な植物を用いて光屈性制御物質の探索を行い、ダイコン下胚軸からraphanusaninsと4-methylthio-3-butenyl isothiocyanate(MTBI)を、アベナ幼葉鞘からuridine, L-tryptophanやavenacoside Bを、トウモロコシ幼葉鞘からbenzoxazolinon類等を単離・同定した。ダイコン下胚軸の光屈性では、光屈性刺激に伴うraphanusanin関連物質の動態と成長抑制活性を精査した結果、青色光照射によって光側組織でmyrosinase遺伝子の発現が誘導され、myrosinase活性が高まり、不活性型の4-methylthio-3-butenyl glucosinolateからglucoseが切り出され活性型のMTBI次いで最も活性の強いraphanusaninsが生成され、光側組織の成長が抑制されることによって光側に屈曲することが強く示唆された。uridineはアベナの他に、トウモロコシやクレスのシュートでも青色光で増量することも分かった。この他、青色光によってその生成が誘導される成長抑制物質として、ダイズからkievitone、クレスから4-hydroxy-2,3-dimethyl-3-nonen-4-olide等を単離・同定した。
