内分泌撹乱物質の環境中運命の解析

研究概要	(1)エストラジオールの環境水中挙動 HPLC-バイオアッセイ検出法を用いて、下水処理水中の女性ホルモン様物質のプロファイリングを行ったところ、主要な物質は天然の女性ホルモン(E1およびE2)であった。一般的な下水処理場におけるE2の除去率は75-88%の範囲であり、他の疎水性汚染物質の除去率と比べて低い成績であった。処理水中の濃度は数ng/L〜数十ng/Lのレベルであり、これは、雄の魚にビテロゲニンを誘導するのに十分な濃度であった。尿中から排泄されたE2の抱合体のうち、グルクロン酸抱合体は下水管の中で分解し、E2へと変化するが、硫酸抱合体は変化せず、そのまま下水処理場へと到達することがわかった。その後の硫酸抱合体の変化についてはさらに研究が必要である。また、E2の底泥への収着特性を調べた結果、線形の収着等温線が得られ、収着量は底質の有機炭素重量比(f_<oc>)と相関関係があることが判った。このことから、通常の環境中に存在する底質については、E2は底質の有機成分に主に収着していると考えられる。実際に、伊勢湾底質中の女性ホルモン濃度を調査した結果、底泥中から検出されたE2濃度は、0〜5.2ng/g、E1濃度は3.4〜12.4ng/gであった。 (2)内因性AhRリガンドの同定アリルハイドロカーボン受容体(AhR)のリガンドによる環境汚染の問題に近年関心が集まり、その対策が焦眉の急となっている。この受容体の真のリガンドと真の役割はまだ不明で、いわゆるオーファン受容体の一つである。我々は、人間の尿中から強力なAhRリガンド、インディルビン及びインディゴを単離同定した。これらの物質はAhRの内因性リガンドである可能性が高い。 (3)人工細胞膜を用いた生物濃縮評価方法の開発内分泌撹乱物質の生物濃縮を推定する方法として、水中からリポソームへの収着量すなわちリポソーム/水収着係数(Klipw)という新しい指標を開発した。今後様々な内分泌撹乱物質でこの指標を測定する予定である。
