新規多連続反応を用いる生理活性多元素環状化合物の効率的合成

研究概要	(4+2)型環化付加反応において1-アザジエンは反応性が低く、適当なジエンとして機能しないことが知られている。しかしながら1-アザジエンの分子内環化付加反応はアルカロイドなどの生理活性物質の合成法として極めて魅力的なアプローチである。検討結果、不飽和アミドをシリル化する反応条件下に多重結合との分子内ディールス・アルダー反応を進行させて多環状化合物を一挙に合成することに成功し、本反応を利用して薬理活性なマピシンおよびルオトニンAの全合成を行った。不飽和アミドと共役不飽和エステル基を併せもつ化合物について連続的に1,4-共役付加反応を行い、橋頭位に窒素原子を持つ多環状化合物の合成法を我々は報告している。本反応をさらに分子間反応に拡張することに成功し、これを用いて抗うつ薬として現在市販されているパロキセチンの簡易合成を行った。分子内での連続する1,4に引き続く1,2-付加反応をさらに分子間反応に展開し、触媒量のルイス酸によって収率良く進行することを証明できた。さらに本手法を発展させて、(4+2)および(2+2)型の環化を連続的に行って、4員環に縮環した多環状化合物の新規合成法を開発した。この反応を利用して最近南米のサンゴから単離きれ細胞毒性を示すパエセレリンAの合成を検討した結果、提出構造の最初の全合成に成功し、このことによって提出構造の誤りを明らかにした。ラジカル反応によってビシクロ[3.2.1]オクタン体を収率良く合成し、これから分子内ディールス・アルダー反応を用いて誘導したカウレン酸メチルエステルが強い神経細胞保護作用を持つことを明らかにした。さらに、炭酸プロパルギルエステルをパラジウム触媒で処理することによって、スピロ融合した環状炭酸エステルを好収率で合成する反応を開発した。この反応は基質に存在する二酸化炭素とフェノール部分が再配列する極めて興味深い反応であり、有用な合成素子を提供する方法である。さらに不斉リガンドを用いることによって、高い不斉収率を持って反応が進行することを示した。
