研究概要	タンパク質は生体内で多種多様な機能を発現しているが、生体外で機能材料として利用するためにはタンパク質の安定性向上、分子間相互作用による階層化構造体の形成等が重要である。本研究では、特に疎水性相互作用に着目したタンパク質の分子設計を行い、特殊環境耐性を有する安定な機能性人工タンパク質材料を構築することを目的として行なった。本研究では安定なタンパク質の創製にあたり、従来行われてきた天然タンパク質の改良という視点ではなく、疎水性結合を導入した頑丈な骨格となる構造部位と、機能を司る機能部位を組み合わせるという設計方針を採った。構造部位と機能部位をそれぞれ一つのユニットと考え、ユニットを組み合わせることによるユニット集合体としてのタンパク質デザインを基本コンセプトとした。さらに分子間での疎水性相互作用により、高次構造体形成を図り、高機能性人工タンパク質材料を構築した。本年度は、前年度に作製したERE-OGPと同様の設計コンセプトに基づき、細胞の増殖を制御する部位としてERE-OGPのOGP部位の代わりにEpidermal Growth Factor(EGF)を配置したERE-EGFも作製した。このタンパク質の基板との接着、細胞接着活性、細胞機能制御活性等を評価した。その結果このタンパク質が細胞培養に有用なマトリックス材料となるとともに、増殖、分化等の細胞機能を制御可能であり、組織構築のためのタンパク質材料としても有用であることが明らかとなった。
