異物、薬物排出ポンプ蛋白の結晶解析と機能

研究概要	病院内感染起因菌の一つである緑膿菌は、構造的にまた作用機作の上からも異なる多くの抗生物質に耐性を示す。この低特異的抗生物質耐性の主たる原因は、この菌がMexAB-OprM異物・抗生物質排出ポンプを発現することに原因している。本研究ではこの異物・抗生物質排出ポンプの作動を明らかとするためポンプサブユニットの結晶回析及び作動機序の研究を行った。ポンプサブユニットの結晶解析に関しては、MexA及びOprMの結晶を取扱った。(i)MexAは元来内膜に結合した膜蛋白であるが、膜結合部位であるN-末の脂肪酸を外したものの結晶化を行った。結晶はPhoton Factory放射光でOscillation angle 1度、カメラ長350mm、100k、distance 350mmでデータ収集を行った。その結果スペースグループP21、格子常数a=90 b=107 c=90Å、解像度3.5Åを得ている。現在のところ重金属置換体を作成中であり、Pt、Os、Sm、Cd、Trなどで安定的なデータが得られており解像度約3.5が恒常的に得られている。(ii)OprMは界面活性剤の存在下に結晶化をすすめており、得られている結果はrhombohedral型R32空間群に属しており、格子常数はa=b=85.24Å、c=1044.96Åである。放射光施設Spring8でのカメラ長350mm、Oscillation angle 1度、温度100k、波長0.9Åでの回析では約2.6Åの解像度を得ている。現在のところ位相を決定すべくHg、Pt及びTh置換体を調整して回析を行っているが、未だ位相の決定には到っていない。この結晶は格子常数c値が高いことから解析の困難さが予想されるが、遠からず構造が解けるものと考えられる。(iii)その他排出ポンプの作動機序の解明のため(a)MexBサブユニットのTMS-4と-10の相互作用の実験、(b)MexEF-OprNポンプの集合体形成及びbiogenesisの実験及び、(c)排出ポンプとDNA-gyrase共役によるキノロン剤高度耐性の機序の解析などを行い各々に成果を得た。
