B細胞分化・多様化を制御する抗原受容体シグナルにおけるBASH/BLNKの役割

研究概要	B細胞特異的アダプターBASHは、Btk, PLCγ2,Vav, Grb2等と結合し、B細胞抗原受容体(BCR)刺激によるPLCγ2やNF-κB活性化に必要である。我々はBASHのSH2ドメインに結合して活性化するHPK1を同定し、HPK1がNF-κB活性化経路を正に制御することを明らかにした。BASH欠損マウスでは、BCRシグナル伝達異常に起因するB細胞の生存・増殖・分化の低下、抗体産生低下などの異常がみられた。本研究では、交配により作製したBASH欠損・抗DNA-H/L鎖knock-inマウスの解析より、BCRのreceptor editingにはBASHが必要であることが明らかになった。さらに、BASH欠損マウスではλ鎖陽性細胞やRS再構成の頻度の低下がみられ、DNA反応性B細胞の割合が増加していることから、receptor editingが正常B細胞レパトアにおいても働いており、自己反応性B細胞の制限に貢献していることが始めて証明された。 BASH欠損マウスではB細胞初期分化が不完全に抑制されていた。一方、BASH遺伝子と同様Pax5の標的遺伝子であるCD19との二重変異マウスではB細胞分化が完全に停止し、蓄積する静止大型プレB細胞上のpre-BCRの発現がさらに増加していた。この細胞ではRAG2が陽性である一方で、κ鎖germline transcriptが減少しており、再構成したκ鎖遺伝子はわずかであった。以上より、プレB細胞の分裂、RAG2やpre-BCRの発現低下、κ鎖遺伝子部位の活性化、さらには小型プレB細胞への分化などを誘導するpre-BCRシグナルにはBASHが必要であるが、CD19がこれらを部分的に補填することが明らかになった。さらに、この二重変異マウスにおいてもH鎖対立遺伝子排除は正常であったが、D断片の読み枠の解析からDμ選択にはBASHが必須であることが判った。従って、両者は異なるシグナル伝達経路により誘導されることが明らかになった。
