IL-18によるTh2サイトカイン産生誘導機構の解明

研究概要	IL-18はIL-12と協力してT細胞、B細胞、NK細胞からIFN-γの産生を誘導する。ところが、IL-18は、IL-12の共存しない場合は、in vitroでIL-2と協力してT細胞からIL-4/IL-13の産生を、また、IL-3と協力して好塩基球と肥満細胞からIL-4/IL-13の産生を誘導する。今回の研究では、この様なIL-18によるTh2サイトカイン誘導に関して研究した。更にIL-18のアレルギー性炎症誘導機構についても検討した。IL-18(5μg)をBALB/c野生型とBALB/cバックのOVA特異的TCRトランスジェニックマウス(Tg)に連日投与した。野生型と同様にOVATgでもIgEの産生が誘導されていた。また、マウスの脾臓から得たCD4陽性T細胞ではIL-4とCD40Lの発現がIL-18投与群で上昇していた。しかし、CD4陽性T細胞を抗CD3抗体と抗CD28抗体で刺激しても、IL-4の産生能を認めなかった。IL-18投与直後から抗IL-4抗体を投与したところ、IgE産生は抑制された。また、抗CD4抗体処理を施したマウスにIL-18を投与しても、IgE産生は認めなかった。以上の結果からIL-18はin vivoでCD4陽性T細胞に作用してCD40LとIL-4の発現を増強してIgE産生を誘導することが明らかとなった。次に、皮膚にcaspase-1あるいはIL-18を過剰発現させたTgを作製し、IgE産生とアトピー性皮膚炎(AD)の発症を検討した。これらのマウスはSPFで飼育されているにも関わらず、高IgE血症を伴う著明なADを呈した。Caspase-1Tgからstat6を欠損させるとIgEは完全に抑制されたが、ADの症状は変化しなかった。ところが、このTgからIL-18を欠損させるとAD症状は消失した。IL-18はIgE非依存的にADを発症させることが証明された。
