IL-18によるTh2サイトカイン産生誘導機能の解明

研究概要	IL-18はIL-12と協力してT細胞、B細胞、NK細胞からIFN-γの産生を誘導する。ところが、IL-18は、IL-12の共存しない場合は、in vitroでIL-2と協力してT細胞からIL-4/IL-13の産生を、また、IL-3と協力して好塩基球と肥満細胞からIL-4/IL-13の産生を誘導する。今回の研究では、IL-18によるTh2サイトカイン/IgE誘導に関して研究した。IL-18(〜5μg)を野生型BALB/cとBALB/cバックのOVA特異的TCRトランスジェニックマウス(Tg)に連日投与した。野生型と同様にOVATgでもIL-18投与量依存的にIgEの産生が誘導された。また、IL-18投与マウスの脾臓から得たCD4^+T細胞はIL-4とCD40Lの発現を上昇していた。IL-18投与直後から抗IL-4抗体を投与したところ、IgE産生は抑制された。また、抗CD4抗体処理を施したマウスにIL-18を投与しても、IgEの産生は誘導されなかった。CD4^+T細胞はNK1.1を発現するNKT細胞と発現しないconventional CD4^+T細胞に分けられる。NKT細胞は構成的にIL-18Rを発現するが、conventional T細胞は発現していなかった。NKT細胞とconventional T細胞をin vitroでIL-2 and/or IL-18で刺激した。IL-2とIL-18で刺激を受けたNKT細胞のみ、強くCD40Lを発現し、IL-4を産生した。更にIL-2とIL-18で刺激されたNKT細胞は、B細胞を刺激して、IgEの産生を誘導した。また、IL-18をCD1欠損マウス(NKT細胞を欠損)あるいはClassII欠損マウス(conventional CD4^+T細胞を欠損)に投与するとIgE産生は誘導されなかった。ところが、classII KOマウスに野生型B6マウスのconventional CD4^+T細胞を移入した後、IL-18を投与するとIgE産生が誘導された。しかし、IL-4KOマウスのconventional CD4^+T細胞を移入した後、IL-18を投与してもIgE産生が誘導された。従って、NKT細胞とconventional CD4^+ T細胞の両者ともに必須であることが明らかとなった。但し、IL-18が標的となるのはNKT細胞で、IL-18で刺激を受けたNKT細胞がconventional CD4^+ T細胞に作用してB細胞からのIgE産生誘導を増強すると考えられる。
