B細胞の抗体産生とアポトーシスを制御する新規遺伝子in vivoにおける機能

研究概要

平成13年度は、CD40Lによって特異的に誘導される新規遺伝子であるClast3とClast5の機能解析を行った。Clast3は264個のアミノ酸からなる細胞質蛋白で、正常B細胞においては、CD40Lをはじめ、抗IgM抗体、IL-4またはLPSなどによってその発現が速やかに誘導されることが判明した。細胞周期での発現は、G0/G1期では低く、S期からM期にかけてその発現が急速に誘導される。NIH3T3細胞でClast3を過剰発現させると、細胞の異常分裂を伴うDNA量の倍数化現象がみられた。逆に、anti-sense oligoを用いてClast3の発現を抑制すると、分裂中期の細胞が減少するとともに、アポトーシスが誘導された。以上の結果により、Clast3は細胞分裂に必須な役割をはたすが、過剰発現すると分裂異常を誘導することが示された。一方、Clast5は、細胞周期のG0/G1でもっとも発現が高く、B細胞の活性化に伴いその発現が消失することが判明した。B細胞株で強制発現させると、細胞周期の進行を阻害することがわかった。また、NIH3T3細胞で過剰発現させると、コロニー形成能を抑制することが示された。従って、Clast5はB細胞活性化に対し抑制的に働くことが示唆された。現在、遺伝子改変マウスを用いた解析を進めているところである。
