痛み刺激受容体と受容体発現神経の可塑性に関する研究

研究概要	TRPV1の制御機構の解明(感覚神経終末レベルでの可塑性)を目指した。炎症関連メデイエイターの1つである細胞外ATPによるPKCを介したTRPV1機能の制御機構を昨年度に報告し、PKCによってリン酸化される2つのセリン残基を同定したが、ブラジキニンによってもB2受容体を介して同様のTRPV1活性化温度閾値の低下が起こることを見出し、この急性炎症性疼痛の発生メカニズムが広くGqに共役した受容体活性化で起こることを証明した。さらに、ATPとTRPV1の機能連関がin vivoで起こっていることを、野生型マウスとTRPV1欠損マウスを用いた行動解析から明らかにした。そして、代謝型ATP受容体のP2Y_2 subtypeが強く関与していることが判明した。発痛物質カプサイシンはまた、逆説的に鎮痛薬としても使われており、その細胞レベルのメカニズムの1つとして細胞外Ca^<2+>依存性の電流減少(脱感作)が推定されている。私たちは、TRPV1チャネルを通して流入したCa^<2+>がカルモジュリン(CaM)と結合してTRPV1に作用して脱感作を起こすことを明らかにし、CaM結合部位を同定した。この部位を欠く変異体では細胞外Ca^<2+>依存性の急性の脱感作が全くみられなかった。この部位に作用する物質は新たな鎮痛薬として機能することが推測される。 TRPV1は痛み受容体であるとともに温度受容体である。現在までにTRPスーパーファミリーに属する5つのイオンチャネルが温度受容体として機能することが明らかになっている。私たちは、TRPV4が35度の活性化温度閾値をもつ新たな温度受容体として機能することを明らかにした。しかし、TRPV4は感覚神経には発現しておらず、視床下部の視索前野の神経や皮膚の角質細胞に発現していた。前者は体温調節中枢として知られており、後者は直接外界の温度にさらされる部位である。皮膚で感知された温度刺激がどのようにして感覚神経に伝えられるであろうか。現在、角質細胞と神経細胞の共培養系で検討を進めている。
