GAL4-UASを用いた時計遺伝子過剰発現システム

研究概要	哺乳類の持つ体内時計においては、特定の遺伝子(時計遺伝子)が約24時問周期で自律的に発現の増減をくり返すことが、リズム発振の中心である。我々は、この時計遺伝子の振動の振幅を増大させ安定化させると考えられる新たな遺伝子群、DBP、HLF、TEF(以上PAR蛋自)及びE4BP4を見い出した。(Genes Dev.2001)体内時計の存在する脳の視交叉上核と呼ばれる部位では、E4BP4は夜間に多く発現し、昼間はPAR蛋白が多く発現していた。さらに興味深いことにE4BP4は時計遺伝子mPer1のプロモーターに結合し抑制するのに対しPAR蛋白は同じ場所に結合しmPer1の転写を活性化する。したがって、共通の結合配列に昼間は活性型、夜は抑制型の転写因子が結合するという極めて巧妙なメカニズムが存在し、時計遺伝子の振幅を大きくしていると考えられる。これらの転写因子の働きを生体内で実証するためGAL4-UASシステムを用いてE4BP4を視交叉上核に過剰発現させたトランスジェニックマウスの作成を行っている。GAL4を視交叉上核に発現させたトランスジェニックマウスは、既に複数ラインを樹立した。UASにE4BP4をつないだトランスジェニックマウスも、複数ライン樹立し、現在GAL4発現マウスと交配中である。今後これらのマウスを形態学的および行動学的に解析することで、E4BP4およびPAR蛋白の生体内での役割を解明する。
