高頻度律動性バースト発火に依存して発現する同期化生成神経回路

研究概要	ラット大脳皮質錐体細胞においては、CaMKIIの活性化によりカチオン・チャネルのCa^<2+>感受性が増大し、その結果、脱分極性スパイク後電位の増強により、高頻度律動性バースト発火(fast rhythmic burst firing, FRB)が引き起こされることを報告してきた。本研究課題に関連して、霊長類研究所と共同研究を行った結果、FRBを示す細胞はサルの前頭連合野において多数見られ、学習課題と関連してバースト発火の頻度を20Hz付近から70Hz付近まで素早く上昇させ、それに伴い、近傍の細胞の活動が迅速に同期化されることを見出した。即ち、バースト発火の周期が高頻度になるに伴い、異なる神経細胞集団間の発火活動のcoherence(同期化の程度)が上昇した。そこで、そうしたFRBニューロンのネットワークを構成し、coherenceがどのように変化するかのシミュレーションを行うために、FRB細胞のモデル化を行い、手始めに少数のFRB細胞間のネットワーク上でシミュレーションを行った。その結果、FRBパターンはカチオン・チャネルのCa^<2+>感受性(K_d値)の増加とともに、doublet、triplet、quartet spikingのバーストパターンに変化していくが、doubletからtriplet、或いはtripletからquartet spikingに変化するK_d値を与えた時に最も、任意の二細胞間において、活動のcoherenceが増加する結果となった。また、相互にシナプス結合をもつ二細胞がsingle spiking modeでランダムに発火し、同期活動は全く認められないが、K_d値を減少させ、doublet或いはtriplet spikingのバースト発火が現れると数サイクルのバースト発火の後に素早く、発火活動の同期化が生じた。こうしたシミュレーション結果は、実際にサルの前頭連合野で観察された結果に極めて類似したものである。神経細胞の周期的活動は、神経細胞群の活動の同期化に必要であり、ある特定の細胞から観察した他の細胞のcoherenceを時空間座標軸に表現した"coherence coding"により、大脳皮質での情報処理が行われている可能性が示唆される。
