運動学習の成立過程における大脳皮質-大脳基底核連関の可塑的神経機構

研究概要

本研究の目的は、運動課題を学習中のサルを用いて、課題遂行時の大脳皮質および大脳基底核の活動変化を経時的に解析し、運動学習の成立過程における「大脳皮質-大脳基底核連関」の可塑的神経機構を明らかにすることである。本研究のもっとも独創的な点は、従来、胸部・腹部疾患の臨床診断などに使用されてきた超音波画像診断装置を脳研究に応用し、超音波ドップラー断層法を用いて課題試行中における脳の血行動態(血流速度の変化)を直接モニターすることにより、神経活動に関連した機能的イメージングを行うことである。平成13年度は、コントロール実験として、片手もしくは両手のボタン押し運動課題を習得したサルから、一次運動野(MI)、補足運動野(SMA)、運動前野背側部(PMd)および腹側部(PMv)など、前頭葉に分布する運動関連領野のニューロン活動を記録した。その結果、(1)MIでは、片手課題で運動開始に応答するような血流増加が反対側性にみとめられた。また、両手課題でも左右の半球で反対側性に運動開始に関連した血流増加がみとめられた。(2)SMAでは、片手課題と両手課題のいずれにおいても、手がかり刺激および運動開始に応答するような血流増加が両側性にみとめられた。(3)PMd(とくに尾側部)では、片手課題および両手課題で、遅延期間に対応した血流増加が反対側性にみとめられた。(4)PMvでは、片手課題および両手課題で、手がかり刺激と運動開始に関連した血流増加が反対側性にみとめられた。以上の所見は、これまでの電気生理学的実験データとよく一致しており、超音波ドップラー断層法による脳機能イメージングが優れた時間分解能と空間分解能を有することが示唆された。さらに、両手運動課題の学習過程において、手がかり刺激および運動開始に関連した血流増加の程度がSMAで特異的に減少するという予備知見が得られており、平成14年度は、このような運動関連領野の可塑的神経機構を中心に詳細な解析を行う予定である。
