研究概要	細胞周期回転による細胞死制御のメカニズムを明らかにするために、我々は、サイクリン依存性キナーゼ(CDK)インヒビターp27^<Kip1>の発現によりB細胞株WEHI-231の細胞死を誘導する系と、p27^<Kip1>の発現により筋芽細胞C2C12の細胞死を阻害する系を樹立し、CDKインヒビターによる細胞死制御が細胞のコンテキストに依存しておこることを明確に示した。さらに、その分子機構を解明するために、WEHI-231細胞に、レトロウイルスベクターを用いて、種々のcDNAフラグメントを導入し、p27^<Kip1>の発現下で生存した細胞からcDNAフラグメントを回収することにより、このような細胞死を制御する遺伝子の同定を行った。同定された遺伝子の1つはc-Myc遺伝子で、そのアンチセンスの発現により、WEHI-231の細胞死が阻害された。さらに、c-MycのアンタゴニストであるMadの発現によっても、細胞死が抑制された。 WEHI-231細胞ではc-Mycの発現が高いが、C2C12では低い。C2C12細胞にc-Mycの発現を誘導すると、細胞死阻害がおこらなくなった。これらの結果は、c-MycがCDKインヒビターによる細胞死制御におけるコンテキスト形成において重要な役割を果たし、そのレベルが高いと細胞死が誘導され、低いと細胞死が阻害されることが明らかとなった。また、細胞分化系として、胎生肝細胞の培養系にて、分化を再現する実験系を確立した。さらに、サイトカインあるい細胞接着による肝細胞の増殖と分化のメカニズムを解明するために、レトロウイルスベクターを用いた遺伝子導入を行い、サイトカインシグナル伝達分子STAT3が種々の肝機能遺伝子の発現に、また、Rasが肝細胞のadherens junction形成に重要であることを示した。
