微小管結合タンパクOrbitによる細胞分裂と減数分裂および細胞分化の制御

研究概要	我々がショウジョウバエから見い出したOrbitは、分子量165kDの微小管結合タンパクである。これが欠損した突然変異体では、著しい染色体の倍数化がおきる。また哺乳類のorthologは、微小管の+端においてCLIP170と結合して微小管の安定性を制御する。チューブリン-GFPを使ってM期の微小管構築をリアルタイム観察した結果、変異体ではアンチパラレルな微小管を束ねる過程に欠損があることを見い出した。新たにそのプロモーター領域にDNAの変異があり、生殖系列でとくに発現量が低下しているhypomorph型変異を分離した。ホモ個体は成虫まで生存できるが、両性とも不妊となる。この雄の減数分裂では体細胞分裂の場合と同様な染色体分配の異常がみられ、細胞質分裂も欠損することを見い出した。これは分裂後期に特徴的なcentral spindleの束が形成されず、その結果収縮環が作られないことによると推測された。体細胞でも細胞質分裂の欠損が観察された。また卵形成の過程において、Orbitタンパクの細胞内局在と変異体の卵巣を調べた結果、Orbitは生殖細胞の形成過程における細胞分裂と減数分裂の進行に必須であり、M期微小管およびfusomeの構築に深く関与することが示された。変異体では、卵母細胞の分化が阻害される。減数分裂終了後、正常な卵室内では卵母細胞内の微小管集合中心を起点とした微小管ネットワークが構築されるが、変異体ではこれができず、BicDなどの卵母細胞の分化に必要なタンパクが卵室内を輸送されないことが示された。Orbitは細胞分裂および減数分裂期における染色体の分配や細胞質分裂の開始あるい間期における細胞内物質輸送に必要な微小管構造の形成を制御している。
