研究概要

本研究の目的は,生物個体の発生過程を制御するメカニズムを明らかにすることである。とくに器官(臓器)の形成にかかわる遺伝子プログラムのダイナミックな展開の様相を明らかにし,ひいては人工臓器や器官再生といった社会的要請にも,基盤的知識や手法を提供しようとするものである。本年度は以下の研究成果が得られた。(1)もっとも左右相称動物の原型に近いと考えられるプラナリアについて,その形態形成と再生に関わる新規の遺伝子がクローニングされ,RNAiによるその抑制は,異所的な脳を誘導することが示された。今後この遺伝子の性質の解明に進むことになる。(2)アフリカツメガエルのアニマルキャップアッセーにより,カルシウム非存在下でアクチビンを作用させることにより,心臓を形成させることに成功した。(3)ショウジョウバエの眼の形成にかかわる遺伝子を網羅的に検索する試みが続けられ,Irx,Smo,Notch,Dackshundなどの関与が明らかになった。(4)ニワトリ胚の肺の形成において,FGF10,Tbx4の重要性が遺伝子導入と実験発生学的手法によって確認された。また腹側に肺の原基が生じることについて,背側では脊索の抑制作用が働くこと,腹側間充織が重要であること,その作用の少なくとも一部はWntllによることが初めて明らかにされた。(5)ショウジョウバエの翅の形成において,形態形成物質であるhedgehogがmaster of thickveinを介して細胞の親和性をコントロールしていることが示された。このように,広範囲の動物種について,器官形成にかかわる遺伝子のカスケードが明らかにされつつある。
