研究概要	発生・再生現象を理解するためには、時間軸に沿った遺伝子発現の詳細を明らかにする必要がある。そのためには遺伝子発現プロフィールの網羅的記載を基に動物種を超えて発生生物学研究に共通に活用されるべき遺伝子発現データベースを構築することに加え,発現プロフィールから遺伝子制御の階層性およびネットワークを予測することを試みることも必要である。第1班においては本年度ホヤcDNAデータベース(東工大・西田)、時間的・空間的遺伝子発現解析を含むアフリカツメガエル頭部特異的遺伝子ESTデータベース(東大・平良)、約70,000配列を収集したESTデータベース(基礎生物学研究所・上野)などについては公開まで進展し、本研究の目的を達成しつつあるとともに、遺伝子を高密度に配列させて遺伝子発現プロフィールを解析するDNAアレイ法を確立するなど、研究基盤の確立において国際的にも大きな貢献をした。現在のDNAアレイ法の問題点として遺伝子発現解析の細胞レベルの解像度(空間分解能)がまだ十分でないことが上げられたが九州大学田代らによってRNAポリメラーゼを用いてRNAを増幅する方法が確立されつつあり、小さな胚を扱う研究者に朗報をもたらした。さらに,網羅的解析から初期発生を制御すると予想された遺伝子やタンパク質の機能を解明するためのハイスループット機能解析系、cDNAライブラリーから形態形成を制御する遺伝子をスクリーニングする発現クローニングが成確立され、興味ある遺伝子が同定されつつある。
