研究概要	研究代表者の島田らは、大腸菌の無細胞タンパク合成系を用い、土壌細菌Parococcus denitrificansチトクロム酸化酵素を機能発現させることに成功した。本酵素の機能発現には、3つのサブユニットI、II、IIIが必須だが、今回これらサブユニットを上記無細胞系で合成し外部から添加した大腸菌の細胞膜に局在させた。Dodecylmaltosideで可溶化後、サブユニットIに付加したヒスチジンタグを利用しNi-NTAカラムで部分精製した試料が、1)3者複合体であること、2)青酸感受性のチトクロムc酸化活性を示すこと、そして3)還元型酵素に特有の可視吸収スペクトル(ヘムaとa_3由来604nmの吸収極大)をもつことを示した。プロトンポンプ活性については、今後の検討課題であるが、上記1)、2)、3)の結果は、本酵素の無細胞系での機能発現を示唆する。分担者の林らは、ミオグロビン(Mb)中のプロトヘムを、非天然ヘムであるポルフィセン鉄錯体(ポルフィリン鉄錯体の構造異性体)に置換し、そのペルオキシダーゼ、ペルオキシゲナーゼ活性を検討した。その結果、この再構成Mbは天然タンパク質よりも酸化触媒として優れていることが実証された。たとえば、グアイアコール(2-methoxyphenol)の一電子酸化の場合、天然Mbに比べ約10倍の加速が、またチオアニソールやスチレンなどの酸素化(二電子酸化)の場合には、約5倍の加速が見られた。また、非天然のヘムとMb変異体を組み合わせると、グアイアコール酸化が約300倍に加速され、天然のペルオキシダーゼに匹敵する。さらに、環境ホルモン、ビスフェノールAをきわめて効率よく酸化分解する触媒として働くことも実証した。また分担者の藤井は、ミオグロビン(Mb)を亜硝酸還元酵素への機能変換を目的とした。亜硝酸還元酵素の配位子であるチロシン残基の役割を明らかにするため、ヒスチジン残基を一つだけ導入したL29H/H64Y-Mb、F45H/H64Y-Mb、L29H/F45H-Mbなどの2重ミュータントを作成した。これらの変異体と合成ヘムの複合体を作成したところ、どれも天然の亜硝酸還元酵素と類似した吸収スペクトルを示した。この結果は、酵素の特異吸収スペクトルがチロシンの配位によると結論した。またこの複合体と亜硝酸イオンは反応することを確認することができ、酵素機能も持ち合わせていることが示唆された。
