味覚・消化器官固有の摂食シグナル連動性水チャンネルの解析

研究概要

小腸におけるAQPの発現様式の解析を以下の通り行う。現在までに、RT-PCRにようて小腸では少なくとも7つ、また、小腸上皮細胞様の分化を示すヒト結腸癌由来の培養細胞Caco-2では少なくとも4つのAQPサブタイプの発現を確認した。
ヒト小腸に発現するAQP分子種についてマウス小腸を用いて、in situハイブリダイゼーション法によって、発現する細胞種の特定を行い、1)AQPO…発現が確認されなかった。2)AQP1…シグナルは絨毛の中心部と絨毛の上皮および陰窩に観察された。3)AQP3…最も強いシグナルが得られた。シグナルは絨毛および陰窩の上皮細胞ほぼ全てに観察された。4)AQP4…陰窩にシグナルが観察された。5)AQP5…十二指腸腺に強く発現していた。6)AQP7…十二指腸腺に強く発現していた。AQP8…発現が確認されなかった。7)AQP9…絨毛および陰窩の上皮の一部にシグナルが観察された。このように小腸にはAQP分子種が多種発現しており、機能的な役割分担を行っている可能性が示唆された。また、十二指腸腺にはAQP1、3、5、7が発現していた。これらで機能分担を行っている可能性が示唆された。さらにAQP9については抗体を用いた詳細な研究を行ったところ、AQP9はゴブレット細胞の側底部の膜上に局在することが判明した。また、AQP9は小腸以外にも、大腸のゴブレット細胞に発現していた。この結果から、ゴブレット細胞が腸管内に粘液を分泌する細胞であることから、ゴブレット細胞特異的に発現するAQP9が小腸および大腸における粘液の恒常性維持に関与していることが示唆された。
