研究概要	1.優性遺伝型式の発症が認められている3家系の腎性尿崩症患者の遺伝子解析を行ない、いずれもC末側のフレームシフト変異であることが判明し、この変異蛋白と正常AQP2蛋白が4量体を形成することを明らかにした・変異蛋白はアフリカツメガエル卵の発現系では細胞内に留まり細胞表面には出現しなかった。 2.培養MDCK細胞を用いて、AQP2をstableに発現する実験系を確立した。通過性のある膜上での培養で、cAMPの刺激によってAQP2が細胞内小胞から管腔膜へ移行することを共焦点顕微鏡ならびに免疫電顕で確認することができた。我々が同定した優性遺伝型式の腎性尿崩症患者に認められたAQP2のC末側の変異を同じ手法で発現させたところ、この変異AQP2蛋白は膜上に発現させた場合にのみ、側底膜に局在しておりcAMPの刺激にも無反応であった。現在変異のどの部分がこの局在の変化に関与しているかを種々の変異蛋白の発現実験から絞り込んでいる。またこの現象をin vivoで再現するためにこの変異遺伝子のトランスジェニックマウスを作成中である。 3.AQP2のC末側に付着し、そのsortingを変化させる蛋白を見つけるため、pulldown法を行ない候補蛋白を同定した。 4.新規のAQP10をクローニングし、水チャネル機能を確認したが、ureaやglycerolは通過させなかった。小腸に主に発現が認められ、小腸での水再吸収に関与していると考えられた。また、既知のAQPについてその詳細な存在部位の同定を行なった。
