水チャネルの多様性のルーツと機能分化の研究:新規水チャネルの機能解析と生理的意義

研究概要	1、ノックアウトマウスの作成:AQPX1はtargeting vectorに入れたものをES cellにelectroporateしたところである。AQPX2はtargeting vectorにいれたところである。AQP10はマウスのゲノム構造をあきらかにしたところである。AQP10は後で述べるようにマウスにおいてはpseudogeneであるのでノックアウトマウスを作成する意義が少ない。 2、AQPX1,AQPX2について培養細胞であるCHO細胞にHAのタグをつけて発現させて細胞内のどの分画にいくかをwestern blotで検討した。細胞膜だけでなくendosomeにも分布していることがあきらかになった。これはendosomeから細胞膜にexocytosisすることで細胞膜のAQPX1、X2の量を調節する可能性を示唆している。 3、AQP10はヒト、ウサギ、モルモットにおいては空腸にのみ発現していた。イヌでは大腸も含めた総ての腸に発現していた。しかし、ラット、マウス、スナネズミではどこにも発現を確認できなかった。脱水や絶食にしても空腸に発現してこなかった。マウスゲノムのORFに突然変異があって意味のあるタンパクを合成できないことからAQP10はげっ歯類ではpseudogeneと考えられた。ヒトでは抗体で空腸上皮の一部の細胞にそまった。アフリカツメガエル卵に発現させると水とグリセリンをよく通した。 4、センチュウに3つゲノム上このファミリーが存在するがPCRでは2つしかクローニングできなかった。ともに水を通すが従来のアクアポリンに比べて非常に弱い。そのうちの1つは抗体で成虫のセンチュウにのみみられ体壁筋かhypodermal cellに発現しているパターンをしめしていた。
