免疫寛容の誘導・維持と破綻における樹状細胞の機能と作用メカニズムの解析

研究概要	免疫寛容の誘導と維持に対する作用ならびにその破綻における役割という両面から生体内における樹状細胞の性状と機能を検討する目的で実験を行い、以下の結果を得た。生体における自己免疫応答が臓器あるいは組織特異的であり、しかもその標的となる抗原は細胞に由来することを考慮し、標識した異系あるいは抗原を取り込ませた自己細胞、腫瘍細胞などを投与し、樹状細胞の捕捉能ならびにそれらに由来する抗原提示能をリンパ器官から調製した樹状細胞を用いて検討したところ、アポトーシス細胞を取り込んだのはCD8^+樹状細胞サブセットであった。また、生細胞を投与した場合には、ホストのNK細胞によって傷害され、その結果CD8^+樹状細胞に捕捉されることが示された。また、組織学的検討によりこれらの細胞はリンパ器官T領域だけでなく、赤脾髄にも分布していることも明らかになった。また、特異的T細胞を用いた抗原提示能を検討したところ、死細胞捕捉能が認められた樹状細胞サブセットにのみ、MHCクラスII及びクラスIを介するT細胞活性化能が検出された。ラテックス粒子や可溶性蛋白抗原に対してはどちらのサブセットもほぼ同じように取り込み、しかもCD4^+T細胞だけでなくCD8^+T細胞をも活性化した。これらの結果から、サブセット特異的な死細胞認識機構の存在が示唆されたと同時に樹状細胞は他の抗原提示細胞とは異なり外来性抗原を内因性抗原としてもプロセッシングできる機能を有することが明らかとなった。そこで、蛋白抗原を負荷したTAPあるいはβ2m欠損細胞を用いて、それらの捕捉に伴う生体内での免疫応答を検討したところ、定常状態における樹状細胞に取り込まれた抗原は特異的T細胞に提示されそれらの一過性の細胞増殖を誘導するものの、エフェクター細胞の誘導には至らず、免疫寛容が成立することが示された。
