ゲノムの死と再生:DNA二重鎖切断とその帰結のバイオロジー

研究概要	A.染色体切断とその帰結。大腸菌で自然に起きる染色体切断を、パルスフィールドゲル電気泳動で検出し、遺伝型と培地への依存性を調べた。染色体の自然切断を遺伝子間の相互の「遺伝的中毒」によって説明した。EcoRI制限酵素修飾酵素の細胞内での安定性を、定常状態および制限酵素修飾酵素遺伝子の細胞からの喪失後に測定した。大腸菌での制限修飾遺伝子のサブゲノム間の転移について、制限酵素活性への依存性を検討した。制限酵素修飾酵素遺伝子の枯草菌染色体上での自己増幅を検出するために、その増幅の迅速なアッセイ系を作り、増幅の引き金になるシグナルを探索した。 B.制限酵素修飾酵素遺伝子とゲノム多型。 Neisseria meningitidisの種内二株のゲノム配列の比較によって、制限修飾遺伝子とISの関与する、ゲノム再編の新しいメカニズムを発見した。 Staphylococcus hominisゲノムの薬剤耐性に関係する動く遺伝子単位の中に、制限酵素遺伝子の痕跡を発見した。 Pyrococcus属の二つの種のゲノム比較などから制限酵素と予想されたORFについて、制限酵素をin vitroで発現した。部分精製し、温度、pH,塩などの反応条件、認識配列、切断様式を決定した。新規の切断様式が示された。 C.制限酵素修飾酵素の命名法。制限酵素修飾酵素およびそれらの遺伝子の多様性の認識に基づいて、それらの新しい命名法を提唱した。
