研究概要	血清中に存在するマンノース結合レクチン(MBL)やフィコリンは自然免疫に働く生体防御レクチンである。本研究の目的は、生体防御レクチンの自然免疫における役割を明らかにするもので、1)MBL-MASP、ficolin/P35-MASPの活性化機構の解明をする。2)フィコリンとMASP,sMAPのノックアウトマウスを作製し、それら分子の自然免疫における役割を解明する。3)ホヤやホヤ同様古典的経路のない円口類のヤツメウナギでのMBL様レクチン、フィコリン、ガレクチン等の構造と機能の解析等を行う。本年度は以下の成果を得た。 1.すでにヒト血清からFicolin/P35を精製し、このFicolin/P35が補体レクチン経路を活性化することを明らかにしてきた。本研究では、Ficolin/P35と類似の構造を持つフィコリン、博多抗原(H-ficolin)を精製し、博多抗原もMASP-1/MASP-2/MASP-3/sMAPと複合体を形成し、補体レクチン経路を活性化することを明らかにした(J.Immunol,in press)。 2.ジーンターゲッティング法により、マウスMASP-1/-3遺伝子の第2エクソンを欠落した遺伝子改変マウス(Masp-1(+/-)と略)を作製し、更にMasp-1(+/-)同士の交配により両染色体に遺伝子変異を持つホモ型MASP-1欠損マウス(以下、Masp-1(-/-))を得た。Masp-1(-/-)はMASP-1とMASP-3をどちらも欠損していた。 3.下等動物におけるレクチン経路の解析:マボヤ体液からフィコリンに相当するレクチンを2種類精製し、そのcDNAのクローニングを行った結果、4種類のフィコリンが得られた(J.Biol.Chem.276,19959,2001)。また、MBL様レクチンGBLがマボヤ補体を活性化し、原始補体系を形成することを明らかにした(J.Immunol 167 4506 2001)。更に、円口類のヤツメウナギにおいて2種類の新規なレクチンを発見し、その構造と機能を解析中である。
