研究概要	網膜の形成過程における前後軸、背腹軸方向の領域特異性の獲得は、後におこる中枢への視神経投射の領域特異性(Topographic projection)の基盤である。我々はE8ニワトリ網膜から両軸方向に発現量の異なる分子の網羅的単離・同定を行う研究を行い、55分子を同定するに至った。 55分子はその構造解析の結果、多くの新規分子を含むとともに、分泌因子、転写調節因子、膜分子、細胞内シグナル伝達分子、酵素等に分類できることが判明した。これら全分子の眼の発生過程における詳細な発現パターンの解析をin situ hybridizationによって行った。この中から、Ventroptinと名付けた、網膜において背腹・前後両軸に対して二重勾配の発現を示す、新規のBMP-4中和因子を発見した。VentroptinはTbx5の発現を抑制し、cVax、ephrinA2の発現を誘導することによって背腹・前後両方向において視神経の投射を調節していることが明らかになった。現在、前後軸方向にはCBF-1,CBF-2とEph,ephrinの間の関係を明らかにしつつある。また、前後軸方向の決定に関与するBMPシグナルの実体を明らかにしつつある。背腹軸方向では、RetinolからRetinalの合成に関わると考えられる酵素の候補分子を同定した。
