網羅的cDNA解析による昆虫個体間・内情報伝達機構の解明

研究概要	本研究の目的は、カイコをモデル動物として、触角、脳、前胸腺、フェロモン腺などの感覚器、神経、内分泌器官由来の平均化cDNAライブラリーを作製し、大量cDNA解析によるESTデータベースを元にした網羅的な解析を行うことで、雌雄個体間で行われるフェロモンを介した情報伝達と、ホルモンによる組織・細胞レベルでの機能調節のメカニズムを分子レベルで解明することである。本年度中に、純系系統p50由来のメスフェロモン腺、オスアンテナ、5齢幼虫神経節のcDNAライブラリーを構築、さらにそれらのライブラリーをすべて平均化した。cDNAライブラリー、平均化ライブラリーから、それぞれからランダムにl-2000クローンのシークエンスを行い、ESTデーター化した。合計で7000個のESTデーターが得られた。フェロモン腺由来ESTからは、これまで単離されていたacyl-CoA desaturaseとは違う新規のacyl-CoA desaturaseが数種類単離された。またフェロモン腺への脂質輸送に関連すると考えられるLow density lipoprotein recepter(LDLR)と有意なホモロジーがある遺伝子が単離され、この遺伝子の発現は成虫では、フェロモン腺特異的、幼虫では翅原基特異的な遺伝子発現パターンを示すことが判明した。さらに輸送された脂質分解に関与すると考えられるリパーゼも数種類ESTから見つかった。これらの遺伝子は、カイコ性フェロモン(bombykol)生合成カスケードに関与するタンパク質群であると推察され、タンパク質の機能解析からの証明を試みている。またアンテナからは、数種類のフェロモン結合タンパク質、感覚器特異的タンパク質群の存在が示され、アンテナでの多様な情報伝達系の存在が示唆された。
