自己免疫性甲状腺疾患の疾患感受性遺伝子の同定

研究概要	甲状腺特異的な自己免疫疾患であるGraves病(GD)と橋本病(HT)が、どのような遺伝要因をそれぞれ特異的にもち、また共有するかを明らかにするために、罹患同胞対法による全ゲノムスキャンと、その結果に基づく相関検定による物理マッピングを行っている。これまでの全ゲノムスキャンによる多点解析により、D8S272近傍で、HT同胞に対してLOD値3.7を示す領域を見出し報告したが、本年度は、この領域約21Mbのゲノム情報から330個の多型性を有するCAリピートをまず同定し、HT103症例、対照群288例を対象にタイピングを行った。カイ2乗検定による相関検定の結果、8q24.2にP値0.00001、オッズ比2.9を示すマイクロサテライトマーカーを同定し、統計遺伝学的にHT発症関連領域を約200kbまでnarrowingした。さらに、narrowingした領域に、ゲノム情報に基づいてgSNPsを数10kb間隔で同定し、同様に、HT103症例、対照群288例を対象にタイピングを行い、カイ2乗検定による相関検定を行った。その結果、P値0.00001を示すSNPを中心として数個のSNPマーカーについて、HTと対照群において有意差が示された。これによって、マイクロサテライトマーカーでの相関が確認されるとともに、統計遺伝学的にHT発症関連領域が数10kbまでnarrowingされた。この領域には、発現が確認されている遺伝子が1つ、遺伝子配列予測プログラムによって遺伝子構造を取る可能性のある配列が2種存在し、また、データ-ベースの更新に伴い、ESTが次々と登録されている。今後、これらを総合的に解析するために、まず、エクソン部分の塩基配列の解析を計画している。また、D5S436近傍に同定された自己免疫性甲状腺疾患全体の発症関連領域約27Mbについても、ゲノム情報から325個の多型性を有するCAリピートを同定しており、自己免疫性甲状腺疾患234症例、対照群288例を対象としたカイ2乗検定による相関検定を行っている。
