海馬神経細胞に高発現する新規高分子量G蛋白質のシナプス関連膜蛋白との機能的協関

研究概要	我々が先にマウス脳の神経細胞において高発現していることを明らかにした新規高分子量Gタンパク質(mOPA1)が果たす機能について、本年は主に培養細胞を用いた発現系にて解析を行った。 COS-7細胞に遺伝子導入したmOPA1は細胞内においてvesicle状の特徴的な局在を示すが、細胞内小器官のマ-カーを用いた共染色により、mOPA1がミトコンドリアに局在していることを明らかにした。mOPA1の過剰発現によりCOS-7細胞のミトコンドリアは形状がtube状からvesicle状と劇的に変化することも判明した。さらに、欠失変異体ならびに緑色蛍光タンパク質との融合タンパク質を用いた解析により、mOPA1のN末端アミノ酸配列がミトコンドリアへの局在を担っていることを明らかにした。また、Gタンパク質としての活性が恒常的に活性化もしくは不活性化しているような点変異体を作製し同様にCOS-7細胞中での局在を観察したところ、作製したいずれの変異体についてもミトコンドリアへの局在が認められたものの、恒常的に活性化したmOPA1変異体を過剰発現した細胞においてはミトコンドリアが核の近傍に凝集する様子が観察され、mOPA1のGタンパク質活性が細胞内におけるミトコンドリア自体の局在に影響を与える可能性が示唆された。なお、初代培養神経細胞の樹状突起においてmOPA1はvesicle状の分布を示すが、必ずしもミトコンドリアマーカーとその分布が一致しないこと、two hybrid法によりGABAA受容体やRGS蛋白を同定したことから、今後神経細胞における細胞内分布と機能的意義をさらに解析していく計画である。
