研究概要	パーキンソン病は老人に多発し、脳内のドーパミン量は加齢とともに低下していくことなどから、老化とドーパミンニューロンとの関連が以前から指摘されているが、その分子メカニズムは全く明らかにされていない。我々は、ドーパミン生合成律速酵素であるチロシン水酸化酵素(TH)の必須の補酵素であるテトラヒドロビオプテリンに注目して、その生合成酵素をノックアウトしたマウスを作製して、ビオプテリン代謝がドーパミン生合成に与える影響を調べた。ビオプテリン生合成の第2段階の酵素であるピルボイルテトラヒドロプテリン合成酵素(PTS)を、相同組換え法により遺伝子破壊した。出生直後のマウスで、Ptsホモ欠損マウスと野生型マウスとを比較したところ、補酵素の欠乏からカテコールアミンやセロトニン量は著明に低下していた。次に、ビオプテリンを補酵素とするTHとトリプトファン水酸化酵素の活性をin vitroで、補酵素添加状態で測定したところ、トリプトファン水酸化酵素活性は全く変化していないのに対して、TH活性は野生型の10%以下に低下していた。THmRNA量には変化が見られなかったが、ウェスタンブロッティング解析から、著明なTHタンパク質量の低下が観察された。また、この低下は特に神経終末で顕著であった。次に、新生仔マウスにテトラヒドロビオプテリンを腹腔内投与して1時間後の脳内ドーパミン・セロトニン量の変化を調べた。ホモ欠損マウスにおいてセロトニン量はビオプテリン非投与群の10倍以上に増加し野生型マウスの約70%にまで回復した。これに対して、ドーパミン量はTHの減少を反映して増加量が少なく、ビオプテリン非投与群に比べて1.5倍にしかならなかった。我々の結果は、老化に伴うビオプテリンの低下により、ドーパミンニューロン内のチロシン水酸化酵素タンパク質量が減少しドーパミン生合成能力が低下することを示唆した。
