Aggresomeの構成成分であるリン酸化αBクリスタリンの生理的・病理的意義

研究概要

平成13年度の研究成果。
(1)ヒトグリオーマ細胞U373MGは恒常的にαBクリスタリンとHsp27が高いレベルで発現している。この細胞にプロテアゾーム阻害剤であるMG-132あるいはラクタチスチンを曝露すると、両分子シャペロンの合成の誘導と、分子内セリン残基のリン酸化ご促進され、それらはとくに不溶性分画に蓄積し、形態学的には核周辺部にアグリゾームと思われる封入体が形成され、その構成成分として、両シャペロンおよびそのリン酸化型とともに、ビメンチン、ユビキチン、プロテアゾーム、βチュブリンが検出された。微小管脱重合促進剤であるノコダゾールを共存させるとアグリゾームの形成は阻止されたが、両シャペロンの合成の誘導、不溶性分画への蓄積、リン酸化の促進は影響を受けなかった。また、リン酸化の促進は、p44/42MAPKおよびp38MAPKカスケードの高進に起因し、その阻害剤であるSB203580またはPD169316によって両シャペロンのリン酸化は阻止された。しかし、合成の誘導と不溶性分画への蓄積とアグリゾーム形成は影響を受けなかった。すなわち、アグリゾーム形成にはリン酸化は無関係であることが明らかになった。HeLa細胞でも、MG-132処理により、Hsp27を含んだアグリゾームの形成が認められた。また、NG108-15細胞に、cystic fibrosisの病因遺伝子を発現させると、αBクリスタリンを含んだアグリゾームの形成が観察された。以上の結果より、αBクリスタリンとHsp27が細胞内蛋白質の品質の管理に関与していることが示唆されるが、それらのリン酸化の意義は不明である
(2)後肢懸垂によって誘導した廃用萎縮ラットヒラメ筋でのαBクリスタリンとHsp27の応答は、上記の培養細胞系でのプロテアゾーム阻害剤曝露時の応答と極似していた。しかも、懸垂中に坐活神経を片側切断すると、切断側の応答が直ちに消滅することから、これらの応答が神経支配を受けていることが明らかになった。
