ブルーム症候群の高発癌性の分子機構

研究概要	ブルーム症候群(BS)原因遺伝子産物の機能を解析するために、高等真核細胞と、ブルーム症候群の相同遺伝子(SGS1)が存在する出芽酵母を用い、以下の諸点を明らかにした。 1.Sgs1とDNAトポイソメラーゼIII(Top3)との機能的関連をTOP3とSGS1遺伝子の二重破壊株を作製し、この細胞に種々の変異をもつTop3やSgs1をコードする遺伝子を導入してその表現型を調べることにより解析した。その結果、top3破壊株の遅い増殖に関わるSgs1の領域はDNA修復に必要なSgs1の領域とは異なること、Sgs1が修復で機能するためにはTop3との相互作用が必要であり、Sgs1・Top3は相同組換え経路で機能すること等が明らかになった。また、Sgs1とTop3との結合に必要な、両者の領域を明らかにした。さらに遺伝学的解析から、Sgs1がDNA複製に関与するDbp11、Sld3、Sld5と機能的関連をもつことを示す結果を得ることができ、BS細胞で観察されるDNA複製の異常を解析するための端緒が開けた。 2.DT40細胞を用いて、BASC(BRCA1 associated genome surveillance complex)中に含まれるタンパク質をコードする遺伝子とBLM遺伝子の二重破壊株を作製して解析し、BLM欠損状態でのSCEの形成にRAD51は関与するが、ATMはほとんど関与せず、MRE11の関与も比較的小さいことが明らかになった。したがって、BLMが欠損した状態で生じるDNA二本鎖切断と放射線によるDNA二本鎖切断では組換え修復の初期過程が異なると考えられる。
