アンドロゲン依存性ラット前立腺癌のアポトーシスにかかわる遺伝子の解明

研究概要

我々が確立し、継代維持しているSV40T抗原トランスジェニックラットは早期より前立腺癌を全例に発症し、去勢によってアポトーシスの過程を経て癌細胞は消失し、萎縮した腺管をみるのみとなる。このラットにtestosterone propionate(TP)をシリコンチューブ(30mg,3週間で入れ替え)にて5週齢から15週齢までの10週間皮下投与し、その後TPチューブの除去と両側精巣摘出を行い、癌の退縮過程を経時的に観察した。この過程において変化をきたす遺伝子を、前立腺腹葉から抽出したRNAを用いてcDNAアレイ、定量的RT-PCRにより検索した。その結果、Caspase 3,6、BAX、Bcl-x、MMP7、TRPM-2、annexin 1,3,4,7の発現が前立腺癌の退縮過程で上昇することが明らかとなった。これらの遺伝子はアポトーシスにかかわるものとして知られているが、本実験により、アポトーシスの過程でその発現は変化し、前立腺癌の退縮と深い関りがあることが示された。これらの遺伝子の働きを解明することにより、前立腺癌の治療に資する有用なデータが得られると考えられる。またこのラットに10週齢から7週間、抗アンドロゲン剤である5α-還元酵素阻害剤のFinasteride(10mg/kg,1週間に5回)、またはアンドロゲンレセプター拮抗剤であるFlutamide(5mg/kg or 20mg/kg,1週間に5回)を投与し、2,5,7週目に屠殺・剖検し、病理組織学的解析を行うと、前立腺癌の部分的な退縮が観察された。抗アンドロゲン剤の効果がこのトランスジェニックラットにおいて明らかに認められたことにより、より効果的な抗アンドロゲン剤のスクリーニングのためのモデル動物としてこのラットが有用であることが示唆された。
