細胞の分化とがん化における大Maf群転写因子の機能解析

研究概要	大Maf群転写因子(MafB、c-Maf、Smaf1、Nrl)の組織形成における機能および発がんにおける関与を明らかにするために以下の研究を実施した。 1.c-Maf遺伝子欠損マウスの死因の解析:c-Maf遺伝子欠損マウスの死因を明らかにするために、c-Mafの発現動態を解析した。その結果、c-Mafは膵臓のランゲルハンス島などの内分泌細胞で発現しており、それらの細胞の機能異常が死因となっている可能性が示唆された。 2.c-Maf遺伝子欠損マウスのc-Maf細菌人工染色体(BAC)DNAトランスジェニックマウスを用いたレスキュー実験:全長150Kbのc-Maf細菌人工染色体(BAC)DNAを有するトランスジェニックマウスを作製し、c-Maf遺伝子欠損マウスのレスキュー実験を行った。現在遺伝子型を解析し、レスキューできるかどうかを解析している。 3.B細胞特異的にc-Mafを過剰発現するトランスジェニックマウスの作製:ヒトの多発性骨髄腫の症例では、その約25%の症例においてc-Maf遺伝子座に微小転座が存在し、c-Mafの過剰発現が観察されることが報告されたが、我々の解析でも多発性骨髄腫の症例でc-Mafの過剰発現が確認された。そこで、B細胞特異的にc-Mafを過剰発現するトランスジェニックマウス作製した.現在までに7ラインのトランスジェニックマウスを作製し、共導入したGFPの発現を確認した。これらのマウスで、多発性骨髄腫が発症するかどうかを解析中である。 4.MafB遺伝子欠損マウスの作製:MafBのコンディショナル・ノックアウトマウスを作製するために、MafB遺伝子の改変ES細胞を樹立した。3回のスクリーニングで5系統の遺伝子改変ES細胞を樹立した。これらの細胞から、マウスの樹立を行う。
