AIKキナーゼファミリーの癌細胞における発現と機能解析

研究概要

Auroraファミリーキナーゼは染色体分配および細胞質分裂に関与すると考えられており、ショウジョウバエのAurora、出芽酵母のIpl1、C. elegansのAIR1およびAIR2、XenopusのEg2などを含む。ヒトにおいては、少なくとも3種類の関連遺伝子(Aurora A、B、C)が存在することが明らかとなっている(課題名にはAIKとしているが、最近Aurora A、B、Cの統一名が提唱された)。ヒトAuroraBについては、dominant negative遺伝子の発現によって細胞質分裂が正常に行われないという報告がなされているが、ヒトAuroraAおよびAuroraCについては細胞分裂時における機能の解析はなされていない。今回、ヒトAuroraファミリーについて以下の諸点を明らかにした。
1)Dominant negative AuroraAのHeLa細胞での過剰発現によって多核細胞が生じ、中心体数に異常が見られた。細胞分裂時には中心体の位置の異常や染色体が赤道面に並ばないなどの異常が見られ、種々の部分欠失GFP融合蛋白質を用いた解析から、Au-roraAのキナーゼ領域中のN-末端領域が中心体への局在に必要であると考えられた。
2)AuroraA及びAuroraBの時間的・局所的発現を詳細に検討し、これらがヒストンH3のSer-10をin vivo及びin vitroでリン酸化することを明らかにした。また、AuroraAが、AuroraBに比べてより効率よくヒストンH3をリン酸化した。
3)細胞分裂の初期に重要な役割を果たすNIMA-related kinases(NEKs)の1つであるヒトNEK7cDNAをクローニングし、その遺伝子がヒト染色体1q31.3に局在することを明らかにした。また、NEK3のcDNAもクローニングし染色体座位を明らかにした。
