膜ミクロドメインのスフィンゴ糖脂質による細胞増殖・分化・死のシグナル制御

研究概要	細胞増殖・分化・死のシグナル調節の場として、GEM、DIMまたはカベオラとよばれるミクロドメインの重要性が認識されてきた。一方、スフィンゴ糖脂質はこのミクロドメインに集積するが、その糖鎖合成酵素遺伝子の操作によって、糖脂質が細胞シグナルを制御していることが多くの系で示されてきた。そこで糖脂質糖鎖のシグナル制御機能につき、糖鎖リモデリング細胞を用いて検討した。GM1を高発現するSwiss 3T3細胞では、PDGF受容体が非カベオラ/GEM分画へ離散し、増殖シグナルが著明に減弱した。PDGF受容体の2量体形成も著明に低下したことより、糖脂質糖鎖の構造がカベオラ/GEMの組成を変化させ、シグナルを調節することが示唆された。また、GM1がPDGF受容体と会合し、そのキナーゼ活性を制御しており、カベオラ/GEM内でのGM1の濃度によりその効果が異なることが示唆された。これらの結果は、抗体を用いた細胞染色の結果からも支持された。一方、Lewis肺癌細胞の実験より、GM2やGM1の新たな発現による転移性とFAKリン酸化の低下など、a-系列糖脂質がインテグリン機能を負に修飾することが示され、インテグリン分子もカベオラ/GEMに存在して、糖脂質により制御されることが示唆された。糖脂質糖鎖の変化によって、細胞増殖因子受容体の局在が劇的に変化したことは、これまでに報告のない知見であり、カベオラ/GEMのシグナル制御の拠点としての位置をさらに高める発見と思われる。また、酸性糖脂質GM1やGD2と受容体やインテグリン分子との会合が証明されたことは、単にこれらがカベオラ/GEM分画に共存するのみならず、物理的に相互作用して機能的に密接な関係を有することを示すものと思われる。今後、これらの分子間相互作用の機構、結合部位、結合による構造変化と機能修飾などの詳細な検討が必要である。
