マトリックスプロテアーゼによるがん細胞の機能制御

研究概要	がん細胞と細胞外微少環境の相互作用はがんの浸潤・転移を決定する重要な因子であり、多様な細胞外プロテアーゼはこの相互作用に大きな影響を及ぼす。本研究では、マトリックスプロテノーゼによるECM分解の生理的意義、およびがん細胞の浸潤・転移への関与について研究を行い、以下の結果を得た。1)上皮基底膜の主要成分であるラミニン5(LN5:α3/β3/γ2)は細胞の接着と運動を効率的に促進する。LN5のγ2鎖N末端領域がメタロプロテアーゼによって限定分解されるとLN5が細胞接着促進型から運動促進型へ移行すること、またγ2鎖N末端部位の強制発現によりヒト膀胱がん細胞のマウス腹腔内での浸潤能が著しく亢進することを明らかにした。これらの結果は細胞の移動や浸潤の調節におけるラミニンγ2鎖N末端部位の重要性を示す。また、新種のLN5アイソフォームであるLN5Bを初めて単離し、この分子がLN5よりもさらに強い接着活性、運動活性、増殖促進活性を示すこと、およびLN5Bもプロテアーゼによる限定分解を受けることを明らかにした。2)腫瘍血管基底膜に蓄積するIGF結合蛋白質様分子であるangiomodulin (AGM/IGFBP-rP1)は弱い細胞接着活性と、IGF/insulin結合活性を示す。今回、ヒト大腸がん細胞にAGMを強制発現させると、腫瘍増殖が顕著に抑制されることが判明した。また、AGMはin vitroで膜結合型セリンプロテアーゼにより限定分解されること、これに伴いAGMのシンデカン1を介する細胞接着活性が増大することが明らかになった。以上のように、がん細胞が産生する細胞外プロテアーゼは周囲のECM分子を限定分解することによってその機能を修飾し、結果としてがん細胞の増殖・浸潤に都合のよい環境を与えていると考えられる。
