炎症とがん:転写因子NF-κBによる抗アポトーシス機構と発がん促進作用

研究概要	炎症起因性を持つフォルボールエステルなどの化合物が発癌プロモーターとしての活性を持つことなどから、炎症と発がんとの関連が注目されてきた。NF-κB(nuclear factor κB)は炎症応答遺伝子群の転写活性化因子として知られるが、発がんとがんの進展にも重要な役割を演じている。NF-κBの主要サブユニットであるp65(RelA)は、がん遺伝子c-Relと同じファミリーに属し、またアポトーシス抑制機能を持つ。我々はp65と結合する因子の包括的遺伝子スクリーニングを酵母two-hybrid screening法を用いて実施し、これらの生物機能の解析を進めてきた。その結果、今年度はp65の転写活性化ドメインに結合する因子として新たに転写コレプレッサー蛋白AES/TLE(Groucho related genes)とTAFII蛋白に属すと考えられるFUS/TLSを同定し、作用機構を明らかにした。コレプレッサーとTAFII蛋白の競合作用の発見は一般的な転写制御の立場からも重要な意義を持つ。また、p65中央部分に結合する因子としてすでに同定した53BP2/Bbp/ASPP2のアポトーシス誘導作用とそのがん細胞株での発現の意義を明らかにし、NF-κBのアポトーシス抑制機構には遺伝子発現を必要としない経路があることを確かめた。今後は、NF-κBを中心とする相互作用因子の同定をさらに進めるとともに、これらの分子間相互作用の全体像を細胞という限られた空間の中で解き、NF-κBによる発がんとがん進展のメカニズムとその弱点を明らかにし、新たながん治療開発の基礎を確立したいと考えている。
