ユビキチン様修飾システムによる細胞増殖制御機構の解析

研究概要	ユビキチンプロテアソームシステムによるタンパク質分解は、細胞周期制御に積極的な役割を担っている。中でもAPC/C(anaphase-promoting complex/cyclosome)とSCFの二種のユビキチン連結酵素は、細胞周期制御において中心的な役割を果たしている。主としてM期の進行を調節するAPCはリン酸化とアダプタータンパク質(Cdc20/Cdh1)の会合によってその活性と標的分子識別が厳格に制御されているが、G1/S期進行において作用するSCF(Skp1-Cullin1-F-box protein-Roc1)複合体の活性制御機構は、ほとんど不明であった。本研究において我々は、SCFのNEDD8修飾がその活性発現に必須であることを明らかにした。NEDD8はユビキチンと58%の相同性を持つユビキチン様タンパク質(UBL:ubiquitin-like protein)であり、全てのCullinファミリータンパク質と共有結合する新規なモディファイヤー分子である。そしてごく最近我々は、SCFがNEDD8修飾によって活性化される分子機構の解明に成功した。即ち、NEDD8修飾されたSCF複合体はE2酵素との結合親和性が上昇し、その結果、E2-ユビキチンのSCF複合体へのリクルートを促進することによってユビキチンリガーゼ活性が飛躍的に増大することが判明したのである。同時に我々は、NEDD8システムが分裂酵母のCullinファミリーの機能ひいては細胞周期制御に必須であること,さらにノックアウトマウス(NEDD8活性化酵素Uba3欠損マウス:早期胎生致死)を用いた解析から哺乳動物でも必須であることを証明し、NEDD8システムの生理学的重要性も明らかにした。
