肺癌特異的E1A発現条件下のNK4遺伝子発現増強による肺癌増殖の抑制

研究概要	我々はE1B55Kのみを欠損した増殖可能アデノウイルスが高い抗腫瘍効果を示す点に着目し、非小細胞肺癌の多くに発現しているSecretory leukoprotease inhibitor(SLPI)のプロモーターによりE1AとEIB19kをドライブするアデノウイルスベクター(AdSLPIE1A)を作製し、SLPIを分泌する非小細胞肺癌特異的な抗腫瘍効果を検討した。in vitro細胞培養系ではSLPIを分泌しない腫瘍細胞にE1A発現を認めず、SLPIを分泌する腫瘍細胞特異的にEIAを発現し、ウイルス粒子増加による感染細胞の増殖抑制を確認した。in vivoにおいてはSLPI分泌ヒト肺腺癌細胞をマウスに樹立し、AdSLPI.E1Aを腫瘍内投与すると有意な腫瘍増殖抑制効果を示した。更にAdSLPI.E1Aにより腫瘍特異的に発現したEIAが、同時に感染した非複製アデノウイルスベクターにtransに作用し、その腫瘍選択的蛋白発現量亢進をねらい、AdCMV.NK4(HGFを介する癌の浸潤・転移における腫瘍間質相互作用をブロックし、かつ新規血管新生抑制作用をもつHGFのN末ペプチドNK4発現アデノウイルスベクター)をこのAdSLPI.EIAに併用し、in vitor、in vivoでの腫瘍抑制効果の増強について検討した。in vitorの系ではEIA存在下によるウイルスの複製とNK4蛋白発現量の増加を認め、in vivoにおいてもマウス皮下腫瘍のモデルに対し単独投与より更に強い腫瘍増殖抑制効果を認めた。以上の結果は、AdSLPI.E1AとAdCMV.NK4との併用によって非小細胞肺癌選択的に強力な抗腫瘍効果を誘導しうることを示している。
