新しい医薬資源からの抗腫瘍性天然物の探索とその分子標的および作用機序の解明

研究概要	血管新生阻害およびアポトーシス誘導療法が新しいがんの治療法として注目されている現在、我々は、海洋生物、カビおよび薬用植物由来の化合物に着目し、多数の化合物に顕著なアポトーシス誘導作用を見出した。まず、オフィオボリンAについては、JNK1-2のリン酸化を促進し、カスパーゼ3の活性化を引き起こし、アポトーシスを誘導することが明らかにされたが、更に、サポニン類のにも同様の効果がある可能性が示唆された。ハレナキノンの構造類縁体であるセストキノンにはアポトーシス誘導活性が認められなかった事から、ハレナキノンの誘導体を種々合成し、PI3キナ-ゼ抑制作用およびアポトーシス誘導活性における構造活性相関を検討する事により、より有用な物質を検討した。真菌由来の化合物であるIC101に、より有用なPI3キナーゼ抑制作用およびアポトーシス誘導活性が認められ、現在、更に詳細に検討している。タイ産生薬マンゴスティーンは、これまで、抗炎症作用が見出されていたが、更にアポトーシス誘導作用が見出された。主要含有化合物であるα-mangostinおよびγ-mangostinにアポトーシス誘導作用が認められ、詳細なる検討により、アポトーシス誘導最終実行分子であるカスパーゼ3の断裂による活性化が認められた。また、抗炎症作用とアポトーシス誘導作用との関連性が報告されており、そのことも踏まえ研究した。がんの治療に関与する血管新生阻害作用を有する事が認められたゴニオドミンAは、細胞接着に対する作用とがんの転移に及ぼす効果が認められた。
